IRF8-/- mice have significantly lower numbers of Ly6C+ monocytes in peripheral organs. BM, spleen and blood from D0, D3, D5 and D7 WNV-infected WT and IRF8-/- mice were processed for flow-cytometric analysis. Inflammatory monocytes were gated as CD45+ CD115+ CD11b+ Ly6G- Ly6C+ cells (a) in the BM (b), spleen (d) and blood (f). Total numbers of Ly6C+ monocytes in the BM (c), spleen (e) and blood (g) were calculated using flow-cytometric percentages and absolute live cells counts for each organ.
