Synthesis of S223-AM. (A) Scheme of synthesis and hydrolysis of S223-AM. The reaction is performed with acetoxymethyl bromide (AM-Br) as a donor of the acetoxymethyl group in the presence of N,N-diisopropylethylamine (DIPEA) in N,N-dimethylformamide (DMF). It results only in the S-linked (S223-AM) but not the O-linked (S223-AM*) version. The postulated mechanisms of hydrolysis resulting in either S223 or OXO are shown. Blue labels, atom identities.
