ASO mismatches differentially affect off-target splicing. (A) MOE-ASOs used to compare the effects of mismatches on off-target splicing include MOE-ASO1 (fully complementary to PKM) and an ASO with five mismatches (highlighted in red). Mismatches at the 3'-end of the ASO have a minimal effect on PKM splice switching (see Supplementary Figure S1). (B) ASO/RNA duplex structures (left) and quantification of the resulting splice change in U87 cells (right). ASOs were delivered by lipid transfection at a final concentration of 30 nM.
