Colons from 12-week-old mdx mice exhibit evidence of a disassembled dystrophin-glycoprotein complex (DGC) and reduced nitric oxide synthase (NOS) activity. Colons from 4-week-old and 12-week-old male C57BL/10 and mdx mice were cut into proximal (P), mid (M), and distal (D) colon segments, lysed, and analyzed by Western immunoblotting for the expression of dystrophin (Dp427) protein (A) and β-dystroglycan protein (β-dys; B). Protein expression was normalized to total protein content as assessed by a total protein stain (TPS). (C) NOS activity was measured in protein extracts from segments of proximal, mid, and distal colon from 12-week-old C57BL/10 and mdx mice using a colorimetric NOS activity assay. Statistical analysis was performed using a 2-way ANOVA with a Bonferonni’s post-hoc multiple comparisons test to determine the effects of genotype and diet.
