Fibronectin is involved in RABV infection. (A) Fibronectin (FN) interacts with ITGB1 and RABV G. Plasmid FN-Flag was cotransfected with plasmid ITGB1-Myc or ERAG-Myc in HEK293 cells at 37°C for 48 h, and FN-Flag interacted with ITGB1-Myc (I) and ERAG-Myc (II) in co-IP assays with HEK293 cell lysates. (B) RGD peptide (GRGDSP) blocked RABV infection in HEK293 (I) and N2a (II) cells in a dose-dependent manner. Different concentrations of GRGDSP were mixed with ERA-eGFP at an MOI of 0.1 at 4°C for 1 h, added to the cells, and incubated at 37°C for 48 h. Dimethyl sulfoxide (DMSO) with the minimum dilution ratio of GRGDSP was used as a control. Relative cell viability and infection were assessed compared with those of the DMSO-treated group, which were set as 100. The data shown are the mean titers ± SD from four independent experiments. The statistical differences were assessed using Student's t test. *, P < 0.05; ***, P < 0.001.
