Tim-3+ cells preferentially upregulate genes that are associated with T cell activation. TGs from acutely infected mice (8 dpi) were removed, processed into a single-cell suspension, and stained for viable cells, CD45, CD8, gB tetramer, Tim-3, and PD-1. CD8+ T cells were gated on gB-CD8+ T cells or Subdom-CD8+ T cells using gB tetramer; Tim-3− PD-1−, Tim-3+ PD-1−, Tim-3+ PD-1+, and Tim-3− PD-1+ cells were sorted, and their transcriptional profile was assessed by using NanoString technology.
