Knockdown of regulated kinases alter osteoblast differentiation. Before induction of differentiation, hMSCs were transfected with siRNAs targeting indicated kinases. Six days postinduction, the degree of differentiation was scored by measuring alkaline phosphatase (ALPL) activity, displayed relative to the scrambled siRNA–transfected control hMSCs: (*) P < 0.05, Student's t-test. Background coloring is based on the observed quantitative proteomics changes during differentiation: (red) decreased protein level; (green) increased protein level; (yellow) altered phosphorylation.
