Effects of podoplanin deletion on anxiety-like behavior. Anxiety-like behavior was evaluated using several tests. (A) In Open Field test (OFT) significant difference in percentage of time spent in the brightly lit center between Pdpn−/− and their wild-type littermate was found (t(16) = 2.29, *P = 0.04, n = 8–10). No significant differences were observed in total distance traveled and the number of entries to the brightly lit center (t(16) = 2.05, P = 0.06, n = 8–10 and t(16) = 1.99, P = 0.07, n = 8–10). (B) No differences were observed in the percentage of time spent in the light area of the Light/Dark Box test (LDB) arena (t(22) = 1.61, P = 0.2, n = 12). Mice from both groups also showed comparable values for the total distances traveled and the number of entries to the LDB arena (t(23) = 1.54, P = 0.2, n = 12–13 and t(22) = 1.16, P = 0.3, n = 12). (C) Pdpn−/− animals presented with significantly less time spent in the open arms of the elevated plus-maze (t(15) = 3.37, **P = 0.005, n = 8–9) and significantly less traveled distance in the open arms (t(15) = 2.16, *P = 0.05, n = 8–9), as compared to their wild type littermates. No significant differences between groups were observed in the number of entries to the open arms (t(14) = 2.08, P = 0.06, n = 8). Data are displayed as mean ± SEM.
