Amphiregulin signaling modulates dust extract (DE)-induced fibroblast wound repair. A circular wound was made in confluent murine lung fibroblast (MLF) (A) or human lung fibroblast (HLF) (C) monolayers, and wells were then treated with medium alone, 5% DE alone, amphiregulin (AREG) alone, or DE + AREG. Alternatively, cells were wounded in the presence of anti-AREG or isotype control antibody (B and D). After 6, 18, and 24 h, the size of the wound area was quantified by image analysis and compared with initial wound areas. Results shown are mean measurements for four independent experiments (n = 16 biological replicates). *P < 0.05 vs. corresponding control time points or indicated comparisons, #P < 0.05 vs. DE, by ANOVA.
