Figure 6

MrgprD is functionally associated with TRP-A1. A) Representative images of β-alanine–induced calcium responses. HEK293 cells were transfected with the expression plasmid of MrgprD (middle) or MrgprD combined with TRP-A1 (bottom). The cells were separately treated with β-alanine (8 mM) and mustard oil (400 μM). The nontransfected cells were used as the blank control. White arrows: β-alanine–responsive HEK293 cells overexpressing either MrgprD or MrgprD+ TRP-A1; yellow arrows: the mustard oil–responsive HEK293 cells with ectopic expression of MrgprD+ TRP-A1. Scale bar, 10 µm. B) Western blot analyses to verify the overexpression of MrgprD and TRP-A1 in transfected HEK293 cells. These cells were of the same transfected batch as was used for the images in A. C) The fluorescence intensities of β-alanine–induced calcium influx in MrgprD-expressing or MrgprD+ TRP-A- coexpressing cells (n = 3). ***P < 0.001, by independent samples t tests. D) Representative traces show β-alanine– and mustard oil–induced calcium responses in nontransfected (left), MrgprD-expressing (middle) and MrgprD+TRP-A1–coexpressing (right) cells.
