Striatin sequestering to cell–cell contact areas occurs prior to junctional maturation. AC) CaCo2 (A) or Huh7 (B, C) cells were treated with 4 mM EGTA in the growth medium for 45 min and then fixed or transferred to normal growth medium for 30 or 60 min before fixation. Fixed cells were stained for occludin or E-cadherin and striatin and examined using a confocal microscope. D) HEK293T and MDCK cells were coimmunostained for actin and striatin. MDCK cells were treated with cytochalasin D (1 μg/ml for 30 min) to depolymerize the actin fibers. Scale bars, 10 µM.
