Nanocapsulated RTX mediates superior B-cell depletion in LNs of rhesus macaques. LNs from three different locations (inguinal, axillary, and mesenteric) were collected at necropsy on Day 21 from Group I rhesus macaques with a single dose (5 mg/kg) of native RTX or n-RTX via IV and processed for immunohistochemical (IHC) analyses. (A) H&E (top) and IHC staining of CD20 (B cell, second), CD3 (T cell, third), and Bcl-6 (Germinal center, bottom) were performed on serial sections. Panels from left to right show the expression level in axillary, mesentery, and inguinal LNs, respectively. Scale bar = 50 μm. (B) 6.8 × 5 cm of each CD20 and CD3 positive staining (n = 7), and 3.4 × 4.2 cm of Bcl-6 (n = 4) were randomly selected and gray pixel values were measured by Image J software.
