Transgene verification. Pancreatic islets and peritoneal macrophages were isolated from 8-week-old WT and Tg mice for the following analyses: A: Transgene PCR. Analyses were performed using cDNA from WT or RIP.iPLA₂β.TG (Tg) mice, and primers that amplify sequence that either spans junction between iPLA₂β and globin. cDNA (200 bp product, lower band) or is within internal control Fabpi (450 bp product, upper band). Lane L, molecular weight ladder; lane N, control reaction without template. B: Insulin content was determined by ELISA using acid-extracted samples.
