(A) Following stimulation with ionomycin and PMA, transcription is activated through the interaction of NFATc2 and AP1 at the ARRE2 element. This is blocked by FK506. (B) Following the expression of Zta, transcription is activated to a lesser extent through an indirect means that is not inhibited by FK506. (C) By combining stimulation with ionomycin and PMA and the expression of Zta, transcription is activated to an intermediate extent. FK506 reduces expression to the level seen when Zta is activated alone. The interaction of Zta with NFATc2 may account for the reduction of the impact of ionomycin and PMA stimulation.
