(A, B) A549 and BEAS-2B cells were transfected with siRNA (50 nM) for 48 h, the mRNA expression of HMGB1 and β-actin were detected using RT-PCR. (C, D) The protein expression of HMGB1 were detected by Western blot after siRNA transfection. (E, F) The siRNA-transfected epithelial cells were treated with TGF- β1 (5 ng/ml) for 24 h, and E-cadherin, Vimentin and HMGB1 expression were detected using Western blot. (G, H) Statistical analysis of the relative expression of E-cadherin, Vimentin and HMGB1 in control and TGF-β1 groups. Data were normalized to GAPDH expression. ##P < 0.01, ###P < 0.001 compared with the value of control group, **P < 0.01, ***P < 0.001 compared with the value of siNC+TGF-β1 group. Data were the representatives of three independent experiments. (I–N) Morphological characteristics of each group were observed under 40× microscope.
