Activation of Gαi2 by MSG plus IMP in isolated muscle cells. Membranes derived from isolated gastric SMCs were incubated with [35S]GTPγS in the presence or absence of MSG (100 μmol/L) and MSG plus IMP (1 μmol/L) for 20 minutes. Aliquots were added to wells precoated with antibodies to Gαi2, Gαs, and Gαgustducin for 2 hours, and the bound radioactivity was measured. An increase in the binding of [35S]GTPγS.Gα reflects activation of G protein. A significant increase in the binding of [35S]GTPγS.Gα complexes was obtained to wells coated with Gαi2 antibody only.
