Expression of Aβ1-42 does not alter levels of tau expression or tau phosphorylation in the C. elegans model. Worms grown to L4 stage were either maintained at 15°C or upshifted to 25°C for 24 h until day one adult stage when populations were harvested. Pellets were approximately 50–100 mg of adult worms (~50 000 individual animals). To assess changes to overall tau expression, blots were probed with a pan-tau antibody (A, representative blot) and quantitated in (B). Aβ1-42;tau-Tg worms were compared to tau-Tg worms; however, no statistically significant change in the level of tau expression was observed. To assess tau phosphorylation, blots were probed with CP-13(ptau Ser202), PHF-1 (ptau Ser396/Ser404) and AT180 (ptau Thr 231) (C, representative blots) and quantitated in (DF). Aβ1-42;tau-Tg worms were compared to tau-Tg worms; however, no statistically significant change in the level of tau expression was observed. Student’s t-test. Error bars represent SEM.
