Their importance is based on the identification of three loss of function polymorphisms of the human P2X7 gene, Q460R, E496A, and I568N (44). Interestingly, the A348 residue (in red on yellow background) is replaced by a T in several mammal and fish species (in blue on yellow background), indicating it constitutes an ancestral variation. In human, the A348T substitution is associated to a major gain of function (45).
