Role of IL-18/IL-18Rα signaling in proinflammatory cytokine production. To investigate the roles of IL-18/IL-18Rα on the production of proinflammatory cytokines, we used ConA (1 μg/mL) and lipopolysaccharide (LPS 2 μg/mL) in the presence of type II collagen (50 μg/mL) for 24 h to stimulate splenocytes in wild-type (WT) and IL-18Rα KO mice (n = 5 and n = 5). Stimulation with ConA and LPS induced the productions of tumor necrosis factor (TNF) and IL-6 as measured by ELISAs. Splenocytes from WT mice with no stimulation were used as controls (n = 5).
