H&E stain and Safranin O stain of knee joint in Dot1l Δ/Δ and Dot1l Δ/Δ; Acan‐CreER littermates. (A) H&E stain of knee joints of control and Dot1l‐deleted mice at 4 weeks of age, 1 week after Dot1l deletion (n = 3). There was disorganization of the growth plate hypertrophic zone and an overall shortening of the growth plate. There was mild decreased articular cartilage thickness without fibrillation or cleft. (B) H&E stain of knee joints of control and Dot1l‐deleted mice at 15 weeks of age, 12 weeks after Dot1l deletion (n = 4). There was persistent marked decrease in articular cartilage and growth plate thickness. (C) Quantification of articular cartilage and growth plate lengths of control and Dot1l‐deleted mice in Fig. 3 A and B.
