TU-inducible changes in the protein expression of ER stress-responsive genes. Distinct cell lines of Nrf1/2^+/+ (A), Nrf1α^−/− (B), Nrf2^−/−ΔTA (C) and caNrf2^ΔN (D) were or were not treated with TU at 2 μg/mL for the indicated times from 0 to 24 h. The TU-inducible changes in the protein expression of differential responsive genes were determined by Western blotting with the indicated antibodies against BIP (as a positive reference, that was also duplicated from Figure 4), p-PERK, p-eIF2α, eIF2α, Chop, p-IRE1 or XBP1. The intensity of blots representing different protein expression levels was then quantified by the Quantity One 4.5.2 software (Bio-rad, Hercules, CA, USA) and shown graphically. In addition, no obvious differences between untreated and DMSO vehicle-treated BIP abundances in Nrf1/2^+/+ cells were also shown (in supplementary Figure S2).
