Biophysical characterization of the truncated pre-miR-1229_SNP GQ sequence. 1H NMR spectra (A) and CD spectra (B) at various KCl concentrations in 10 mM cacodylic acid, pH 6.5, demonstrating the formation of a GQ structure. The UV thermal denaturation hypochromic transition (C) at 10 µM RNA in 150 mM KCl was fit using Equation (1) (Materials and Methods), to determine a Tm of ~78 °C. The Tm values plotted as a function of the RNA concentration (D) revealed that an intramolecular GQ structure is formed. Nondenaturing gel electrophoresis (E) visualized by UV shadow (left panel) and stained with the GQ-specific NMM dye (right panel), revealing the formation of a GQ structure at all KCl concentrations.
