LGK974 inhibits renal cancer cell proliferation and induces apoptosis. (a, b) Colony formation assays: LGK974 (10 μM) were applied to 786-O and ACHN cells for 96 h. (c, d) 786-O and ACHN cells were stained with Annexin V-FITC/PI and analyzed via flow cytometry. LGK974 (5 μM or 10 μM for 786-O cells and 10 μM or 15 μM for ACHN cells) acted on the two cell lines. (e, f) Western blotting experiments were performed by treating renal cancer cells with DMSO, at different concentrations of LGK974 (786-O, 5 μM or 10 μM; ACHN, 10 μM or 15 μM) for 72 h and then proteins were extracted. The protein levels of Bax and Bcl2 were determined via immunoblotting. All data are expressed as the mean ± SEM. The experiment was repeated at least three times. Statistical significance was determined using two-tailed Student's t-test or one-way ANOVA. p < 0.05; ∗∗p < 0.01; ∗∗∗p < 0.001.
