Figure 3. — Deletion of the SE region reduces HLA-DRB1 and HLA-DQA1 mRNA levels. RNA was isolated from two independent isolates for each designated mutant line and subjected to qRT-PCR for expression of the indicated MHC-II gene or CIITA. WT represents duplicate Raji cell cultures. Data were normalized to the percentage of 18s rRNA, and the mean ± SD is shown. Each culture was analyzed three independent times, and significance was determined by two-tailed Student’s t tests between the WT and other samples. *, P ≤ 0.05. A one-way ANOVA with Tukey’s post hoc test gave similar P values when sample groups (WT, Δ21, Δ18) were combined. No significant differences between samples under the bar labeled ns were observed.
