P_HpFMD enables strong derepression and exceeds the strength of methanol induced endogenous P. pastoris promoters. Strains bearing selected promoters from Fig. 1 (HpFMD, HpMOX, PpAOX1, PpCAT1, PpGAP) were cultivated in shake flasks and inoculated to a low starting OD₆₀₀ of 0.05. Reporter protein fluorescence, OD₆₀₀ and glucose levels were measured. Fluorescence/OD₆₀₀ values at t = 0 are not shown, as the starting OD₆₀₀ of 0.05 was outside the linear range of the spectrometer used.
