Inhibition of O-GlcNAc prevents mice from DSS- and AIEC LF82-induced intestinal inflammation. C57BL/6 mice were given 3% DSS in drinking water and/or 2 × 108 AIEC LF82 per day in 200 μL PBS by gavage for 2 weeks. Mice were simultaneously treated with 1 mg/kg of DON dissolved in 200 µl PBS by i.p. on day 1 to day 3, then administered every other day until the end of the experiment. (a) The body weights of the mice were monitored every day. (b) Survival of the mice. (c) Colon intestine epithelia of the mice were fixed for the O-GlcNAc staining by IHC at the end of experiment. Scale bar = 50 µm. (d) Percentage of samples with high or low expression of O-GlcNAc in (c). (e) H-scores of O-GlcNAc in (c). * P < .05 and ** P < .01 (Student's t-test). (g) Illustration of O-GlcNAc in the development of intestinal inflammation induced by AIEC LF82.
