Figure 2

Elevated IOP results in a progressive reduction of dACs and RGCs in the GCL. A,B: Vertical sections of control and bead-injected retinas were labeled for Brn3a to identify RGCs and counterstained with a nuclear dye DAPI to differentiate dACs. C: By eight weeks after initial microbead injection the total number of cells in the GCL (DAPI-positive), total number of RGCs (Brn3a-positive), and total number of dACs (difference of DAPI- and Brn3a-positive cells), were all significantly reduced from control levels. D,E: Histograms quantifying cell counts performed at weekly intervals show a statistically significant decrease in the numbers of both dACs and RGCs beginning at 4 weeks after microbead injection and a further reduction at 8 weeks. Bars indicate mean value ± SEM. *** p<0.001; Student’s t-test. Scale bars =50 μm (applies to panels A and B).
