A and B, The given cell lines plated in 96-well plates were exposed to the indicated concentrations of VRT752271 (VRT) alone, erlotinib (Erl) or afatinib (Afa) alone or their respective combinations. After 3 days, cell numbers were estimated using the SRB assay. The data are means ± SDs of four replicate determinations. The numbers inside the graphs are CIs for the given combinations. C and D, PC-9/AR cells were exposed to DMSO, 0.5 μM VRT or 2 μM GDC, 100 nM the tested EGFR-TKI or their respective combination for 48 h and then harvested for detection of apoptosis with annexin V/flow cytometry (C) and for detection of protein cleavage with Western blotting (D).
