Inhibition of protein arginine methyltransferase 1 (PRMT1) with AMI-1 reduces serum-induced activation of cultured renal interstitial fibroblasts in a dose-dependent manner. Normally cultured NRK-49F cells were treated with AMI-1 (0–10 μM) for 36 h. Cell lysates were then prepared and subjected to immunoblot analysis with antibodies against α-smooth muscle actin (α-SMA), collagen type I, PRMT1, asymmetric di-methylated histone 4 arginine 3 (H4R3me2a), or GAPDH (A). Collagen type I (B), α-SMA (C), PRMT1 (D), or H4R3me2a (E) were quantified by densitometry and normalized with GAPDH.
