Inhibition of EGFR increased TLR4 induced tumor-associated inflammation. CAL27 cells were cultured in 12-well plates, pretreated with 100 μg/mL cetuximab or gefitinib for 2 h, and then stimulated with 1 μg/mL LPS for 24 h. Supernatants were collected and analyzed for cytokine TNF-α (A), PGE2 (B) and NO (C) production using ELISAs. (D) The expression of PGE2 and NO synthetase was detected by western blotting using anti-COX2 and anti-iNOS antibodies.
