The reaction scheme was drawn following the experimental results reported in this work and previous studies.21, 24 The nucleophilic Sec residue of reduced TGR could be covalently modified by electrophilic aromatic inhibitors (A). The modified C-terminus moves to FAD redox center of the other subunit (B). The electrophilic aromatic ring of bonded inhibitor accepts a single electron from the thiol of proximal C154 and then transfers it to oxygen molecule to produce superoxide (CE). EWG = electron withdrawing group.
