TGR (500 nM) was incubated with NADPH (100 μM) and compounds for 30 minutes. The inhibitor concentrations were 20 μM for 1, 2, 58 and 50 μM for 3 and 4. Samples were then desalted to remove unreacted compounds. NADPH consumption, superoxide production using pyrogallol red, and H2O2 production using Amplex Red and horseradish peroxidase were determined. Addition of 10 units of SOD abolished oxidation of pyrogallol red and addition of 20 units of catalase inhibited oxidation of Amplex Red (not shown).
