Localization of TDO2 RNA and TDO protein in hepatic metastasis specimens (Case 15 and 16). In situ hybridization of TDO2 RNA (ac,fh): Positive cells with TDO2 RNA show a distinct red stain. Metastatic uveal melanoma cells show stronger and distinct staining (c,h) compared to the surrounding liver (b,g). Staining with unrelated control probe is shown as negative control in the mid-left inserts. TDO protein staining (d,i): Areas positive for in situ staining of TDO2 RNA are also strongly positive for TDO protein. Staining with isotype-matched control antibody is shown in the mid-left inserts. HMB45 staining (e,j): The areas strongly positive for TDO2 RNA (c,h) and TDO protein are also positive for HMB45. Neighboring liver tissue with much weaker staining for TDO2 RNA (b,g) and TDO protein is negative for HMB45. Staining with isotype-matched control antibody is shown in the mid-left inserts.
