Sensitivity limit and versatility of polysome profiling in SW55Ti tubes. (a) 1–8 OD_{260 nm} of HEK-293 lysates were loaded onto 10%–50% sucrose gradients (in SW55Ti specific tubes) and centrifuged at 45,000 RPM for 65 min at 4 °C (left). Peak readability was compared to the one of conventional polysome profiles made in SW41Ti tubes, where 5–20 OD_{260 nm} were loaded onto 10%–50% gradients which were centrifuged at 35,000 RPM for 3 h at 4 °C (right). (b) Sucrose gradients of varying sucrose concentration ranges were prepared in SW55Ti tubes (a–f). Profiles in b + e and c + f were shifted up by 110 and 200 mV, respectively, to prevent overlapping of curve traces (to aid interpretation).
