Induction of HO-1 inhibits PRV replication. (A,B) PK-15 or (C,D) ST cells were inoculated with PRV at a MOI of 0.01 for 1 h at 37°C, followed by treatment with 0, 10, 25, 50 μM of CoPP. Cells and supernatants were harvested to detect HO-1 and gB protein expression via western blotting, and measure supernatant progeny virus titers using TCID50, at 36 hpi. (E) Indirect immunofluorescence assay was performed. (F,G) PK-15 cells were transfected with 100 nM of siHO-1 or siNC for 12 h, then cells were infected with PRV at a MOI of 0.01, in the presence or absence of 50 μM of CoPP. Cells were harvested to detect gB and HO-1 mRNA and protein expression levels via RT-qPCR and western blotting at 36 hpi, respectively.
