Effect of Mettl3 knockdown on nuclear export of Traf6 mRNA. (A,B) The protein levels of TRAF6 in the Mettl3-shRNA and Mettl3-shCtrl RAW264.7 cells were detected by Western blotting after RANKL stimulation for 0, 15, 30 and 60 min. β-Actin was used as an internal control. (C) The total mRNA expression of Traf6 was assessed using qRT-PCR in the Mettl3-shRNA and Mettl3-shCtrl groups. (D) The distribution of Traf6 mRNA in the nucleus and cytoplasm was analyzed by qRT-PCR. The data are presented relative to the levels of β-actin (cytoplasm) or U6 (nucleus).
