Role of p53 in reovirus replication. (A)–(D) A549 cells were transfected with a plasmid expressing p53 or siRNA targeting p53 and then infected with MPC/04 at an MOI of 5 after 24 h of transfection. Twenty-four hours after transfection, the relative expression of the viral S4 gene was analyzed by real-time PCR (A and C). The expression of p53 was examined by western blots (A and C). The virus titers were measured (B and D). (E) and (F) Twenty-four hours after transfection with siRNA targeting p53, A549 cells were inoculated with Nutlin (20 μM) or DMSO before infection and then infected with MPC/04 at an MOI of 5 in the presence of drug for 24 h. The relative expression of the viral S4 gene was analyzed by real-time PCR, (E) and the cell supernatants were harvested for an analysis of the virus titer (F).
