(A) Serum-starved mIMCD3 cells and (B) serum-starved NIH-3T3 cells were incubated for 5 min with C5a or IL-6 in the absence or presence of the respective RTK antagonist AG1478 (for mIMCD3, 30 μM) or PDGFR tyrosine kinase Inhibitor III [PDGFR-inhib](for NIH-3T3; 10 μM) after which the cells were assayed for Src, Akt, and Erk phosphorylation by Luminex assay. (C) Serum-starved mIMCD3 cells and (D) serum-starved NIH-3T3 cells were incubated for 1 h with C5a (100 ng/ml) or IL-6 (10 ng/ml) in the absence or presence of the respective RTK antagonist AG1478 (for mIMCD3, 30 μM) or PDGFR tyrosine kinase Inhibitor III [PDGFR-inhib](for NIH-3T3; 10 μM), after which the cells were extracted with TRIZOL and Bcl-2, Bcl-xL, Bim, and Bax mRNA transcripts quantitated by qPCR.
