293T cells expressing the wild-type or the mutant FIV Env glycoproteins together with HIV-1 Tat protein were dissociated and equivalent numbers of cells were added at 1:5 ratio to 4×104 TZM-bl cells. Coculture was continued for 48 h, after which cells were stained for β-galactosidase and scored for syncytia formation. A fraction of the transfected 293T cells was subjected to immunoblotting to monitor Env expression.

Data presented were obtained from three independent experiments, and each assay was performed in triplicate. Blue foci were quantitated as described in “Materials and methods”. Values represent means±standard deviations.
