Total lysine content of axonal proteins. Protein reactivity of 1,2-DAB and formation of high molecular weight adducts correlated with lysine content. Both in vivo and in vitro, rat neurofilament (NF) heavy chain (NF-H) and NF-M formed polymers more readily than either NF-L or tubulin. Other 1,2-DAB protein targets involved in anterograde axonal transport included rat microtubule-associated tau, the rat motor proteins kinesin and dynein, and the rabbit glycolytic enzymes glyceraldehyde phosphate dehydrogenase (GAPDH: 8% lysine) and lactate dehydrogenase (LDH: 7–8% lysine).
