Plaque reduction assay. After transfection of a virus-specific siRNA at 1 and 10 nM concentrations, cells were infected with different dilutions of Coxsackievirus B3 (CVB3). The infection with a virus dilution of 10−5 of 10 nM transfected cells targeting CVB3 resulted in fewer visible plaques than for the 10−6 dilution of the virus alone. The CVB3 titer was reduced more than one order of magnitude. Mock: uninfected cells, siCtr.: control siRNA with no matches to either the viral or target organism genomes.
