To quantify the staining of permeabilized cells, cells were first fixed with 3.5% paraformaldehyde for 25 min, permeabilized for 10 min in 0.25% Triton X-100 or 0.5% octylglucoside (Calbiochem) and non-specific binding sites blocked with 10% horse serum for 15 min on ice. As negative control for this staining, an anti-Coronavirus S protein mAb was used (Co).
