DC838 could inhibit the mouse spleen cell proliferation dose-dependently. Spleen cells isolated from naive mice were cultured with 5 μg /mLof Con A for 72 h in the presence of increasing concentration of compounds: •, CsA (0.01, 0.1, 0.5, 1, 2, 5, 8, 10, 100 μM); ▴, DC838 (0.01, 0.1, 0.5, 1, 2, 5, 8, 10, 100 μM). After culture, MTT assay was used for measuring the cell viability. Each value indicates the mean ± SD of three experiments using 3 mice with triplicate sets in each assay. Half-maximal inhibitor concentrations (IC50 values) are obtained by log-probit analysis of inhibitory curves using SPSS, a Windows statistical software package.
