A and B) Three longitudinal sera samples from a single asymptomatic blood donor were tested at the indicated times for the presence of IgM (A) and IgG (B) antibodies against rNS1 antigen of USUV and WNV. C and D) Detection of IgG (C) or IgM (D) antibodies from 44 sera samples of blood donors by rNS1-based ELISA assay (EI and FD for commercial Eurimmun and Focus Diagnostics, respectively). Optimal cut-off value of WNV P/N ratio for IgG and IgM fell at 1.7 and 1.4, respectively, and was extended also to USUV for the analysis. Each ELISA result includes the average of two biological replicates. Results are shown as the average of two biological replicates, single values are indicated by symbols on the graphs. Cutoff threshold as determined in the text is indicated by the dotted line.
