Conformational diversification of Wisc-1 prions in deer expressing PrPC polymorphisms Ser-96 and His-95. Structural stability of deer PrPCWD after exposure to increasing concentrations of GdnHCl (m). The apparent fractional change (Fapp) of unfolding was measured by CDI with 8G8 or 12B2 mAbs before or after proteinase K treatment (PK −/+). CWD conformers encoded in all three PrP primary structures were detected with 8G8 mAb, whereas 12B2 only recognized prions encoded on WT–PrP. Black dotted lines indicate the denaturation midpoints (GdnHCl1/2). Fapp values and bars represent the mean ± S.E. obtained for each individual deer CWD lineage (two batches of 20% w/v brain homogenate) measured in triplicate with each Eu-labeled mAb.
