DNA vaccines encoding consensus H7 proteins elicit broad HAI antibody responses in mice. Sera from each group of immunized and unimmunized mice were serially diluted by two-fold with PBS in a 96-well polystyrene microtiter plate with 25 μL in each well. A portion of 25 μL virus suspension containing 4 hemagglutinin units (HAU) H7N9, H7N7, H7N3, and H7N1) were added to each well. After incubation of the plate at room temperature for 1 h, 50 μL of 0.5% (v/v) chicken red blood cells were added to each well, and the plate was incubated at room temperature for 30 minutes. HAI titers were determined as the highest serum dilution that completely inhibited hemagglutination. Values are the mean titers plus the standard error of the mean (error bars) (A–F).
