Poliovirus binds low-molecular-weight GlcNAc oligosaccharides. (A) Preparation of low-molecular-weight GlcNAc oligosaccharides. Chitin was hydrolyzed by acid to generate water-soluble oligosaccharides. GlcNAc oligomers were fractionated with a Bio-Gel P-4 size exclusion column and analyzed by FACE. Left lane, oligo(GlcNAc) standard. (B) Poliovirus binds to GlcNAc oligomer-coated WGA-agarose beads. 35S-labeled poliovirus (105 PFU) was incubated with glycan-coated lectin-agarose beads. After incubation, bead-associated virus was measured by scintillation counting. WGA lectin binds GlcNAc-containing glycans; ConA binds glucose-containing glycans.
