Effect of exogenous PGE₂ added to differentiating wild-type MM6 cells. MM6 cells were differentiated with TGF-β + 1,25diOHvitD3 (VD3) and increasing concentrations of PGE₂ (1 to 10 µM) present during the 96-h differentiation period. In addition, cells were differentiated with TGF-β, 1,25diOHvitD3, and zymosan, ±dexamethasone (Dexa, 150 nM). Whole-cell lysates were analyzed by Western blot (WB) using Dicer antibody (Ab) 1701 to 1912. The membrane was reblotted with Dicer Ab 600 to 650 and with mPGES-1 Ab. Similar results were obtained in two additional experiments.
