Phenotypic analysis of amiR-β1/β2-cop exposed to salt (NaCl). (A) Wild type (Col-0) and amiR-β1/β2-cop seeds were sown on 0.5 × MS for control conditions and 0.5 × MS supplemented with 100 mM NaCl and 150 mM NaCl in Petri plates. The percentage of seedlings with green cotyledons was calculated after 12 days. Data are mean ± s.e.m. (n = 100) of four independent experiments. (B) Seeds of wild type (Col-0) and the amiR-β1/β2-cop mutant were sown on MS plates without salt and grown for 4 days before being transferred to MS plates containing 160 mM NaCl. Three days after transplantation, the percentage of seedlings with green cotyledons was calculated and expressed as mean ± s.e.m. (n = 60) of three independent experiments (right panel). An image of a representative experiment is shown in the left panel. (C) Ion leakage of wild type and the amiR-β1/β2-cop mutant 1 day after transplanting seedlings to MS plates containing 135 mM NaCl.
