A) Effects of PSL and TO-207 on activated CD8+ T cells. Peripheral blood CD8+ T cells (2 × 104/well) were activated with anti-CD3 and anti-CD28 mAbs (Dynabeads human T-activators CD3 and CD28), and treated with PSL or TO-207. Supernatants were recovered after 72 h. The error bars represent standard errors (SEs) from three independent experiments. B) Effects of PSL and TO-207 on CAR T cell cytokine production. K562/CD19 cells (1 × 104) were co-cultured with CD4+ or CD8+ CAR T cells (5 × 104). The cells were treated with different concentrations of PSL and TO-207. Supernatants were recovered 24 h later. The error bars represent SEs from three independent experiments. The linear dose-response relationship was assessed using log-transformed dose values (to the base 10) in a mixed model, in which the zero dose was replaced by the log (minimal dose) - 1. P < 0.05 was considered statistically significant. n.s.: not significant.
